An insightful presentation by Dr. Dirk A. Lamprecht on “Targeting de novo Purine Biosynthesis as a Therapeutic Strategy for Tuberculosis.”
The presentation focuses on targeting the de novo purine biosynthesis pathway in Mycobacterium tuberculosis, an approach that has gained significant attention in recent TB drug discovery research.
Dr. Lamprecht is an author of a 2025 Nature study reporting the discovery of JNJ-6640, a first-in-class small-molecule inhibitor targeting PurF, the first enzyme in the mycobacterial de novo purine biosynthesis pathway. The study found that JNJ-6640 showed nanomolar bactericidal activity in vitro and demonstrated proof-of-concept efficacy in vivo using a long-acting injectable formulation.
The research provides evidence for targeting a previously underexplored metabolic pathway in tuberculosis drug discovery and highlights the potential of PurF inhibition as a strategy for developing new antituberculosis therapies. The study also examined the activity of JNJ-6640 against drug-resistant tuberculosis and its potential role in future treatment regimens.
