Prokaryotic Single-Cell Transcriptomics for Understanding Staphylococcus aureus Infection Heterogeneity and Immune Evasion

🔴 One of the featured presentations during Session 4 (Day 2) of the 2nd International Virtual Conference on Staphylococci and Staphylococcal Infections 2026 featured Dr. Lee Korshoj from the University of Nebraska Medical Center (USA), who delivered an insightful lecture on Prokaryotic Single-Cell Transcriptomics for Understanding Staphylococcus aureus Infection Heterogeneity and Immune Evasion.

Although bacterial populations are often studied as uniform communities, individual Staphylococcus aureus cells can exhibit striking differences in gene expression, metabolism, and stress responses during infection. This cellular heterogeneity enables subpopulations of bacteria to adopt specialized physiological states that enhance survival under antibiotic treatment and immune pressure. Conventional bulk transcriptomic approaches measure the average behavior of millions of cells, often masking these important differences. Recent advances in prokaryotic single-cell transcriptomics now allow researchers to examine gene expression at the level of individual bacterial cells, providing unprecedented insight into the diversity that exists within a single infection.

During his presentation, Dr. Korshoj discussed how single-cell transcriptomic technologies are transforming our understanding of S. aureus infection biology. The lecture demonstrated how these approaches can identify distinct bacterial subpopulations that differ in their physiological state, revealing mechanisms that contribute to immune evasion, persistence, and infection heterogeneity. By resolving transcriptional variation at single-cell resolution, this research provides a more detailed understanding of how bacterial populations adapt to changing host environments and respond to antimicrobial and immune challenges.

Dr. Korshoj’s research focuses on bacterial gene regulation, infection biology, and the development of advanced genomic technologies to investigate host-pathogen interactions. His laboratory integrates molecular microbiology, microbial genomics, computational biology, and single-cell sequencing approaches to study how phenotypic diversity within bacterial populations influences virulence, persistence, and antimicrobial tolerance.
